GLE Modulates Testicular Injury, DNA Damage, PCNA, Ki67 and P53 Alteration in PTZ Induced Epileptic Seizure Disease in Rat

Abstract
Background: Pentylenetetrazole (PTZ) is one from the most drugs to develop epileptic seizures disease. Pentylenetetrazole develops chemical kindling and it is reported that PTZ-kindled seizures mimic. Epilepsy is the term of the human brain neurological disorder characterized by seizures affecting people of different ages, race, social class or geographical borders. Objective: The aim of the present study was to investigate the possible prophylactic effect of ginkgo leaf extracts (GLE) on the testicular injury, PCNA, Ki67, P53 alterations and DNA damage in PTZ -induced epileptic seizures disease in rat. Method: Forty healthy male albino rats weighing 140 ±10g and of 9-10 week’s age were used in the experiments, rats were divided into four groups (1st group was control group which never received any treatment. 2nd group was positive control that animals were treated with GLE (100 mg/kg body weight), 3rd group was PTZ group that rats were treated with pentylenetetrazol (PTZ; 40 mg//kg body weight) once per day every other day for nine days, 4th group was post treated PTZ with GLE group). Result and conclusion: PTZ induced testicular injury as disturbance and abnormal distribution of spermatocytes in the seminiferous tubules with marked degeneration and a significant decrease in sperm numbers with presence of many syncytial cells. Also, PTZ induced DNA damage, significant decrease in PCNA and KI67, and an increased in P53 expressions in testicular tissues. Treatment PTZ with GLE improved testicular injury, DNA damage, increased PCNA and KI67 and decrease P53 expressions.
